Biochemistry 302: Lineweaver-Burke plot


What is a Lineweaver-Burke plot?
It's a graph of the inverses of velocity (Vo) and substrate concentration [S] of a reaction. Velocity (Vo) and substrate concentration [S] can be measured experimentally. The graph plotted from their inverses gives straight lines. Where the line crosses the 1/Vo axis is 1/Vmax. Where the line crosses the horizontal 1/[S] axis, is -1/Km. The uninhibited enzyme makes a flatter line.

What information is easily obtained from such a plot?
Vmax and Km.
